Solvent Delivery
LC-20A solvent delivery pump systems cover a wide application range with optimised performance under any condition. LC-20A series consists of 3 pump types: The tandem piston pump, LC-20AT, the dual-piston pump, LC-20AD and the binary dual-piston pump, LC-20AB. In terms of durability and performance the LC-20A series pumps are a further improvement of the proven LC-10AVP series providing a number of additional useful features and improvements. As part of an HPLC system those pumps can incorporate the CBM-20Alite interface to control the entire LC system via web browser or instrument control software. Prominence series solvent delivery pumps can be equipped with optional automatic piston washing kit. The gradient valve for LC-20AD and LC-20AT can be used as well for solvent selection and be combined with binary or ternary high pressure gradient systems.
The LC-20AD and LC-20AB pump designed in micro volume double piston technology provides pulsation free solvent delivery from real micro flow applications up to semi-preparative flow rates. Merely select the suitable mixing chamber for the appropriate application range. The flow rate ranges from 0.1 µl/min up to 10 ml/min. The high resolution flow rate stetting combined with the small piston stroke volume allows for gradient elution in semi micro range without using an additional flow splitter. The pumps can be configured as quaternary low pressure gradient system (AD) and as high resolution (0,1 %) binary- or ternary- high pressure gradient system. Beside this dual independent high pressure gradients can be operated simultaneously in one HPLC system.
Within an HPLC system 2 LC-20AB can be used for independent binary high pressure gradient elution. Beside this it is possible to combine up to 4 LC-20AB pumps to simultaneously perform the same binary high pressure gradient elution, which enhances configuration flexibility of multiplexed systems.
The LC-20AT with serial piston design, which covers the range from semi micro to semi preparative flow ranges, can be configured as quaternary LPGE as well as binary- and ternary HPGE system. To minimise the internal dead volume the low pressure gradient valve is installed into the pump unit. This robust pump for routine application provides high flow rate accuracy within the flow rate range from 1 µl/min up to 10 ml/min.
Sample Injection System
The SIL-20A/AC is using a high resolution metering pump and the direct sample injection principle for sample injection, which means no loss of sample for the injection process. Even with smallest injection volumes the metering pump resolution of 6 nl provides excellent precision and accuracy. The autosampler can be operated via the Communication Bus Module, CBM-20A / Lite (or by SCL-10AVP System Controller in compatibility mode) as well as in stand alone mode. The module SIL-20AC is equipped with a powerful Peltier Sample Cooler which allows for keeping the sample temperature down to 4° C.
SIL-20A/20AC is capable of handling small injection volumes from 0.1 ml up to 100 ml (2 ml Option) and combines fastest injection speed (10 s for a 10 ml injection) with excellent precision, accuracy and linearity. The autosampler can handle various vial types ( 1, 1.5 or 4 ml vials) as well as microtiter plates (96-well, 96-deep-well or 384-well).
Nearly complete elimination of sample carry over is achieved by the following construction details:
Direct sample injection principle, in which the sample suction line is part of the flow line.
Special needle coating
Minimised contact surfaces of thoroughly selected materials.
For very critical samples the optional second rinsing pump allows further reduction of sample carry over.
To control the entire HPLC system using a chromatography data system or web browser the CBM-20Alite interface can be installed into the SIL-20A/AC.
Rack Changer
With the optional Rack Changer using 12 microtiter plates on 4 stacks sample capacity can be increased to 1152 plus 10 standard vials. Each of the 4 stacks can hold up to 3 microtiter plates or deep well microtiter plates. Different autosampler needle stroke settings allow for parallel use of microtiter plates and deep well plates.
During operation stacks which are currently not processed can be removed and filled with microtiter plates, which increases work efficiency.
The powerful Peltier elements and the well designed isolation of the Rack Changer/C type allows for sample cooling and heating from 4° -40° C.
Combining the Rack Changer with an injection speed of 10 seconds per injection and the excellent LC-20 series gradient pumps makes Prominence an ideal system for Fast HPLC and LC/MS/MS.
Compared to common autosamplers with typical cycle times from 30-60 seconds, SIL-20A will provide at least 5 hours economy of time when running 12 microtiter plates
Column Thermostat
prominence column ovens - CTO-20A and CTO-20AC can accommodate multiple columns, mixing chamber, two column switching valves as well as two manual injectors. The fan allows equal temperature distribution inside the whole compartment. The temperature control range for the CTO-10A extends from RT %2B 10°C to 85° C and for the CTO-20AC the temperature control range extends from RT -10°C to 85°C. Additionally Prominence column ovens have the capability for temperature gradient programming. A built-in ready state sensor with flexible ready range and wait time settings assures that samples are only injected after the temperature has been completely stabilised.
For tracing the column history a Column Management Device (CMD) can be used and connected directly to the column oven. The high pressure switching valves, FCV-12AH and FCV-14AH, can be directly controlled by the column oven. The actual valve positions of the high pressure switching valves are displayed on the column oven display. Constant analytical conditions can be achieved due to the possibility of accommodating all required modules and accessory, e.g. an FCV-12AH 6-port valve for automatic sample pre-treatment with trap column.
An environment temperature sensor allows for exact and precise temperature control of all column ovens. The column ovens can as well be used in stand alone mode and controlled via the build in keyboard.
HPLC-Detectors
prominence UV-VIS Detectors
SPD-20A/AV UV/VIS detectors have expanded linear range (up to 2,5AU) and highest sensitivity also in dual channel mode. The extremely low baseline noise and the excellent wavelength precision allow to achieve constant high precision chromatographic analysis.
Even if the SPD-20A/AV is operated in stand alone mode, the complete functionality such as wavelength programming or wavelength scans is applicable using the build in keyboard. SPD-20A with built-in D2 lamp covers a wavelength range from 190-700 nm. The SPD-20AV UV/VIS detector with built-in D2- and Tungsten-Halogen lamp covers a wavelength range from 190-900 nm.
An improved optical system and higher-order digital filter increases the signal to noise ratio, improves the wavelength accuracy, wavelength reproducibility and expands the wavelength range of the successful previous models.
The thermostatic flow cell improves baseline stability especially when using mobile phases with absorbance that fluctuates easily due to temperature changes.
The above listed improvements lead to the result that the SPD-20A/AV has the lowest baseline noise and baseline drift among the currently available HPLC UV/VIS detectors.
Maintenance works are simplified because of easy access to the detector cell and pre aligned lamps. Build ion validation functions, as automatic wavelength accuracy test, a built-in Mercury lamp for wavelength calibration, maintenance - and error- logs supporting work according to GLP/GMP. Simply by changing the detector flow cells the UV/VIS detectors can be customised for all kind of applications from micro LC up to preparative LC.
HPLC-Detectors
prominence Photodiode Array Detector
The SPD-M20A is the newest product out of a long experience (since 1982) developing photodiodearray detectors. Offering a very low baseline noise of less than 0.35 x 10-5 AU it is comparable or rather more sensitive as common UV detectors. The SPD-M10A is supposed to be the ideal tool for spectral identification and library searches in trace analysis by an optical resolution of 1.2 nm and spectra range of 190-800nm. Variable slit width (1.2 or 8 nm) allows an adaptation according to the application of use. Fast Scan periods and digital filter of higher order lead to a fast detector response and a high data collecting frequency (40Hz).
The SPD-M20A can be used for applications from the micro-LC up to preparative LC by changing the flow cell. The thermostated flow cell stabilizes the baseline and avoid temperature depend effect in eluents which absorbance is temperature dependent. As for an multi-wavelength UV detector, the SPD-M20A offers a 4 channel analogue outlet. The 4 channels can be imported to other data processing system or can be used for as triggers for fraction collection.
The prominence PDA detector communicates with the network / PC via an Ethernet interface, and is controlled by LabSolution software. The software package allows beside the standard data processing features and instrument control for complete prominence HPLC systems peak purity calculation, library searches and flexible reporting. The integrated web-server allows easy access to the main parameters with Microsoft Internet Explorer®.
Maintenance work is minimised by simple access to the detector cell and lamps from the front of the instrument. pre-adjusted D2 Lamps avoid time consuming adjustments of the instrument after lamp exchange. The software supports wavelength tests by inbuilt Holmium filter, energy test for optimisation of the data acquisition and logbook functions in order to meet GLP/GMP requirements.
