This high performance 2D capillary-to-microflow HPLC system co-developed by Shimadzu and the NIH performs separation of complex peptide mixtures with a level of sensitivity and resolution that meets or exceeds present 2D gel electrophoresis. Combining cation exchange chromotography in the first dimension with capillary reversed-phase chromatography in the second dimension, and using a trap column that allows for various separation conditions to be optimised independently, the system can analyse large numbers of trace peptides from digests.

It has proved to be an extremely effective tool for the discovery of unknown proteins with low expression levels. When combined with a high performance mass spectrometer with an electrospray ionisation interface (ESI), unequalled performance and sensitivity is achieved.
